CD4+and CD8+T cells of an HLA-B8+donor recognize different EBV latent antigens. Unseparated, CD8 or CD4 depleted, T cell enriched PBMCs were stimulated for two weeks with autologous LCL. Afterwards the cultures were tested for their CD4/CD8 content by flow cytometry (top row). Their antigen specificity was tested using autologous dendritic cells infected with recombinant vaccinia viruses encoding EBNA1, EBNA2, EBNA3B, EBNA3C, LMP1, LMP2 and BMLF1 (B1) in IFN-γ ELISPOT assays. As a positive control reactivity against the autologous LCL (LCL) was measured. 104 expanded T cells were added per ELISPOT well. One representative of three experiments is shown.
